EN Structural information is crucial for understanding biological systems at the molecular level. Herein, we describe examples of Medicago truncatula proteins studied structurally by protein crystallography, that are related to hormonal regulation by either being a part of the signal transduction cascade, or being able to bind phytohormone molecules. The chapter summarizes previous work on histidine-containing phosphotransfer protein 1 (MtHPt1) and two examples of pathogenesis-related proteins of class 10 (PR-10), namely Nodulin 13 (MtN13) and Phytohormone-Binding Protein (MtPhBP). Another structure, of histidine-containing phosphotransfer protein 2 (MtHPt2), is reported here for the first time, together with a description of the methodology that proved effective in obtaining high amounts of highly pure M. truncatula proteins.
